Would you consider this a reasonable offer for UP?
Hello,
Newby question here. I got an offer yesterday from QR for DOH - ISB (a little over 3 hour flight) and it is $442. A friend of mine who is flying two weeks from now, same flight got an offer of $255.
What is reasonable here? Does QR drops the offers as it gets closer to the date of flight?

It would be neat if your enquiry produced a range of prices paid for upgrades on the route at different days-to-departure. QR likely has that information included in the wider data on fares for that route - but all you can hope for is anecdotal evidence and unhelpful comments.
QR's data resources should allow it to pitch offers at a level that will generate the level of response it wants. The marketing team might follow the path of a Dutch auction until that response is achieved, but that's not really a trajectory full-service carriers want to get involved in. In general they'd prefer to avoid the situation where on-line forums could with confidence advise passengers to hold back a purchase because the price will inevitably fall.
Here's my anecdotal and likely unhelpful comment: having decided my budget allows only for Y-travel, I'd rather spend $400+ upgrading my garden than go against that initial decision in order to gain a nicer seat for a three hour trip. Fortunately for airlines, there will be others with wildly different but perfectly valid views on how best to employ their cash.
Your list of loyalty programmes memberships suggests you'll be well looked after on the ground whatever your class of travel.

Me too - I would rather spend $400 in my garden also then for a three hour flight.
