Aleli Cottages Culebra PR

Hello all. First time post so go easy on me.
I booked direct through hotel to Aleli Cottages who has a nonrefundable deposit policy. I canceled 22 minutes later but never heard back from them and I did not follow up. Now more than one week later they charged me the deposit. When I contacted them again after the charge they referred to their policy and sent me a cancelation email. I think they missed my initial email which is when I should have received the cancelation email. I asked the manager 4 times if they saw the initial email and she never answered once.
It seems deceptive for them to send the cancelation email one week after the cancelation request and only after the deposit was charged to my account. Thoughts?
Do hotels have certain time frame that they have to respond to inquiries?
How do I speak to the owner and not just the manager who doesn't want to admit her mistake.
Does anybody think I have a case or I am SOL.
