Miss one flight in middle of booking
Some friends are flying the folllowing route:
MAN - FRA
FRA - BKK (5 nights)
BKK - SYD (7 nights)
SIN - FRA
FRA - MAN
All with Lufthansa
Separately they booked SYD - SIN (5 nights in SIN) with Qantas.
They no longer need to go to SYD but would like to go to PER instead. Their ticket is non-changeable and non-refundable. They are happy to separately book BKK - PER -SIN and would like to cafch the remainder if their itinerary. Is there any possibility not to fly BKK - SYD and not lose the remainder of the Lufthansa flights??

Unless they contact the operating airline to rebook they would be deemed a no show and the remainder of the ticket would be cancelled.

As the operating and perhaps ticketing carrier is LH, it is possible that if the tickets were purchased in Germany, that your friends might have tickets where not all segments must be flown to avoid cancellation. It s extraordinarily unlikely because such tickets are generally very expensive and not likely wholly inflexible as you say theirs' are.
Nonetheless, it is always worth reviewing the specific fare rules for the specific tickets just in case theirs are of the sort which would permit them to drop a segment.
The likelihood however is that as these are inflexible tickets, that they will not be able to pay a fee and make a change. Rather, they would need to count on having their tickets cancelled when they no show for a segment and having to purchase new tickets in order to continue.

Best way to avoid problems is to leave this ticket as is and fly all segments. Then buy a separate SYD-PER-SYD. If the SYD-SIN flight on QF is flexible / changeable can modify it to PER-SIN. A bit extra flying back and forth and some loss of time, but least risk to lose money / return flights.
