Arriving Incheon @ 4:30am from US
We have a trip planned next year at the end of August right before Labor Day. Lodging is still TBD but assuming somewhere in Seoul central to attractions and sights. Our outbound itinerary as follows:
SFO -> ICN: Friday 11/25 11:30pm - Sunday 11/27 4:30am (13h travel) on Asiana booked via an award ticket with Air Canada.
Question: With our arrival at 4:30am on a Sunday, is there anything special to make note of regarding transportation to Seoul or getting entry into the country at the airport or anything else in general?

Arriving 4.30am IS early but airport railway runs from 5:23am on sundays so no prob to get into the city. (?????)
there is direct service to Seoul station, as well as commuter service. if you choose latter then you will need Korean Won to buy T-money card (much like metro card) from a vending machine. the direct train tickets can be bought with CC as well as cash from a information desk.
Travel time will be about 40mins to Seoul station, from there you can transfer to Seoul metro to get to your final destionation/hotel.

the train is also the forward/backward-facing 2/2 seat arrangement with luggage racks up top (as opposed to the commuter service having the standard Metro-style center-facing seats along the sides)

This is a late reply but my travels are for later this year in August (not April), so we're still in the planning stages. Likely plan to use the express and store luggage at our final destination. Not sure if they'll allow that early of a check in, but we still haven't booked accommodations yet so TBD.
