when do changes in Medallion status become effective?
I'm Delta Gold now, today I'll qualify for Platinum in 2020. Is it effective tomorrow or on 1 Jan 2020?

You will be X for the rest of this Medallion year and then that will continue from 1 Feb 2020 thru 31 Jan 2021

It's already afternoon and you're using future tense, so I'm guessing that your qualifying flight is late today. Hence I would expect miles for the flight to post tomorrow and your status to be updated the following day, Sunday. It will be effective at the time it shows in your DL account.
In the past, people apparently needed to remove and re-enter the FF number on existing bookings when status increased, but more recent reports have indicated that this isn't necessary. Still, if you'll be in the middle of a short RT, I wouldn't count on upgrades being prioritized at the higher status right away.

You will upgrade the day after the qualifying activity posts, not when you fly it.
