Point reservations usually carry the same restrictions as a cash reservation. Some properties allow cancellation up to 6pm the day of check in, some have a 3-day cancellation policy. This should show on the web page before you confirm the reservation. If you do cancel the reservation, the points will be returned within a day.
One thing to note is if you cancel, the points will go back into the same expiration bucket from which they came. In the olden days this was a good way to extend your points by making a reservation and then cancelling it, the points were returned to the current bucket. But alas, another hole filled in... :(
