Is this itinerary possible on 2-4-1?
Hi,
Would the following itinerary be possible on an Amex 241 as a single booking :
LHR-MIA-ORL-MIA-PLS-LHR
Trip involves Disneyland and Turks & Caicos, the latter trying to take advantage of the very low Avios needed for MIA-PLS
I would consider different routing but don't see how that's possible at the moment!

No.
Your 2-4-1 is only valid on BA flight numbers flown by BA planes.
So you can use the 2-4-1 LGW-MCO (thereby avoiding MIA immigration) and PLS-LON, then either make a separate redemption booking or a cash booking MCO-PLS.

Is this itinerary possible on 24?
Just looked and MCO is ex Gatwick which is a shame as wanted to use CCR on F redemption.
Also can't book Avios MCO-PLS it seems. MIA-PLS is 4500 avios e/w in economy, bargain!
Might be more hassle than its worth with my itinerary

Unless AA fly MCO-PLS, you would have to go via MIA anyway, so 9000 Avios pp.
The CCR is overrated, especially in this instance and assuming seats are available on both flights - you can save 34k Avios going straight to MCO (as you pay extra for the flight up to MCO from MIA), miss out on the variable that is MIA immigration, get a good few extra hours on your holiday etc etc.
With the points you save (fees will be about Ł25 lower each, too), find somewhere at Gatwick for a lavish champagne breakfast. Valuing those saved Avios at 1p each, you could readily get a bottle of Dom at the Caviar house and a balik salmon breakfast for two, and still come out well ahead *before* you consider the time savings.
