Amex has special relationships with fine dining restaurants - where they reserve tables for centurion card holders. Some concierge specialists have used their pull to get this tables for platinum card holders. You can request it by asking nicely for them "to do everything they can" to get the table.

Amex plat got me a reservation to French Laundry. Dinner reservations, only about 40 days out (reservations start 60 days out). No availability online. I was very happy with this.

Being the procrastinator I am, I forgot about mother's day brunch (don't tell her). I called the concierge up and gave them two places I like, and they got me into one of them. The even better part was that this was for a party of 6, so not easy even on a regular Sunday!

So far I have had a couple of good experiences with the concierge. The first one was getting tickets for Cirque du Soleil O in Las Vegas, calling in the morning for the show that same evening. The second one was just last month, when they got me a reservation at Le Bernadin, in New York, with about ten days notice. And later they were able to add an extra person to the party, completely last minute.
Since I'm quite of a football fan I regularly call them about tickets, but find them consistently expensive, due to always resorting to their 'alternative' providers. No luck so far in that area with them.

I had a recent positive experience with a non-standard request:
I have an upcoming short trip and wanted to hit a few popular high-end bars/lounges type places that require you to be seated (fairly limited capacity) but dont take reservations and waits can apparently be horrible at certain peak hours. I asked the concierge if they could find out what times the places get hit with long waits.
Sure enough, a few hours later I was emailed a detailed email explaining what each establishment reported re: peak hours, with a note reminding that one of the places was cash only.
Happy that they can do more than basically act as a human Opentable ^

I needed to have a piece of siding replaced on my house - concierge called every distributor in a 100 mile radius of me to find one who had the exact pieces I needed in stock and would sell individual pieces (as opposed to a case).

Costco Delivery
Concierge coordinated moving a large sectional couch from Costco 20 miles away to my apartment in the city.
They called 3 delivery companies, got quotes and summarised my options. Two of the delivery shops wanted $300-400. Amex Concierge found a fantastic one for $150 ^

I tried to get tickets to the Statue of Liberty. I know they are sold out far ahead, but Amex didn't even make any effort. Just told me to call hotels, because they just buy them off the website, too and hotels might have "something".
