This was handled as it ought to have been by the Captain. I presume that he made arrangements for the FA to not be involved in service for the micro-hop and, having done so, would have written up the incident.
OP, if he wishes, could send a short note to AA. AA will send that to Mesa for action, although it is more than likely that Mesa will already have dealt with the issue.
DOT won't bother with a rude FA issue. All it will do is forward the complaint to AA who will, in turn forward to Mesa.
AA may toss a few miles OP's wife's way for the inconvenience, but don't expect some massive review to be turned over.

This is the correct response. Their EAP is a good possible solution. It sounds like it could be personal issues affecting her work performance

Years back a GA at LGA came onboard and let our every cuss word known to man to the F cabin. The FAs looked aghast. The Captain came out of the cockpit and ordered her off the a/c. I wrote AA and got a nothing response. There she was still working flights,

This seems like a typical FA on many of my AA and affiliated regional carrier flights these days. Horrible, just horrible. Some are very good, but many are not and the experience is so inconsistent that inconsistency is the only consistent thing about AA these days.

If I were in charge at AA, I'd have such GAs terminated on the spot and escorted by airport police directly to the outbound subway station.

On the contrary for me at least all I've encountered have been very pleasant and professional flight attendants ...PSA, Envoy, or Mesa.

Who is hiring these people? I have encountered a similar LUS mainline FA out of PHX and it's baffling to me. If I acted like this in my job I would have been fired years ago.

Maybe it's me, but asking for any coffee drink as a PDB strikes me as unnecessarily high maintenance. The FA doesn't brew coffee just in case someone in F orders it during boarding. They have to make a pot, then dump it before takeoff. How do you expect them to play Candy Crush if they're serving passengers during boarding.
