Yes to Canaima, and I saw Isla Margarita from the bus 
list of towns/cities:
Caracas
Barcelona
Cumana
Laguna de Tacarigua (to see the 1000's of red Ibis)
Maturin
Temblador
(then a visit to some Warao natives on the river)
the beautiful historic city of Ciudad Bolivar on the Rio Orinoco
Canaima lagoon
boat to Angel Falls, camping overnight
all in all a great visit in 2 weeks (7 friends flew SFO-CCS on AA for the trip)
