Standby requests for revenue passengers are still dependent on manual entry by a CSR (BP5A/5B depending on the situation), but employee travel BPs are assigned automatically. Actually - things are a lot better now that employee travel *has* gone electronic (we call the program eTravel). Manual entry of boarding priorities at check-in has gone the way of the dinosaur, as each traveller along with their electronic travel authority has the BPs listed in the authority by operating carrier/system.

Upon check-in, the ACI system cross references the employee number with the respective travel authority, assigns the correct boarding priority/seniority date and adds them to the DM list automatically. Absolutely no need to assign manually unless it's an employee of a non-UA/UAX airline, in which case it's still a paper ticket anyways.

In response to the OP - if your original flight was cancelled, and you were on standby as a BP5B (other revenue standby), you would be entitled to a BP5A (disserviced passenger) standby priority upgrade, but you would have to make contact with an airport CSR to make this change.
