If you are not looking for non-stop or direct, you can get single through DL fares to/from PTP from many US cities. It's just that these through fares will require overnight layovers in MIA because of morning departure from MIA to PTP and the return flight arrival in the evening. Fares to PTP allow layovers of up to 24 hours as do most international fares (other than Canada/Mexico). While AF MIA-PTP flights may have had a stop at PAP in the past, these are now non-stop flights.

DL has 3 daily non-stop flights between MCO and MIA and MCO-MIA-PTP is a valid routing for DL MCO-PTP through fares. However, the earliest MCO-MIA flight does not arrive in time to catch the same day MIA-PTP flight. So you will need to book the MCO-PTP direction the day before the MIA-PTP flight and layover the night at MIA. There's no way around this. In the reverse direction, the last DL MIA-MCO flight is late enough in the evening that you can book PTP-MIA-MCO routing on same day. The screenshot of Google Flights I posted above reflects these routings and fares. However, there are no MCO-ATL flights available after the MIA-MCO evening flight arrives, so you will have a layover overnight in MCO on the return to ATL.

As far as cheapest DL fares on US to PPT, I'm 99.99% sure the $445 MCO-PTP RT fares are it (again, these are through fares even though you connect in MIA). While you can't check from every possible origin in US, I checked from a number of competitive US gateway cities (plus a number of smaller DL stations) on Google Flights and none of them were cheaper. The only ones that were close were out of BOS and JAX at $463 roundtrip.

If you are just looking for cheapest fares to PPT regardless of carrier, JetBlue looks like it wins here with 3x weekly non-stop flights on JFK-PTP starting in November from $318 roundtrip (for Blue Basic fares, $358 for Blue).

The fact that the AF flight between MIA and PTP leaves in the morning and arrives back in the evening imposes a lot of limitations. If you want 100% DL tickets, you will need overnight layovers in both directions. There's no way around this. If you are willing to book other carriers to reposition from ATL to/from MIA, and then book AF MIA-PTP RT on a separate ticket, you can potentially avoid overnight layovers. The problem with booking separate carriers is that you will potentially lose protection in the case of IRROPS. The absolute cheapest option looks to be repositioning to JFK and flying B6 to PTP. I doubt you will get any cheaper than this by island hopping on one of the Caribbean carriers to PTP.
