I can confirm that I was able to buy a comfort upgrade ZRH-LCY on a ticket issued by our corporate travel agent. I did online after the TA had issued the ticket before.
(Note. It is only possible after TA has isseued the ticket, which sometimes takes some days...

I alsonoted that on some of my booked flights flights the option disappeared after some days - turned out these were the flights that were literally full
