Personally, I would not recommend the Hilton Molino Stucky or the JW for a first trip to Venezia. I'm not saying that they are bad properties, just that I would stay on the main island for the experience of it. Nothing beats simply walking out the door and wandering the maze of passageways as opposed to waiting for a boat to shuttle you anywhere,.
