Emphatically, yes. Anyone in boarding group 5 has to stop by the ticket counter, where they'll be asked to check any bag that appears to be too big to qualify as a personal item.
The gate agents will mention "no full-size carry-on for Group 5" as part of the boarding announcements; I don't know that I've ever seen them have to check a bag, though, presumably due to the ticket counter interaction.
