From first principles, I think that the answer to this must be no. This is because on each flight in the booking, all the passengers in the booking must be booked into the same booking class on that flight.
However, if 2 pax are booked into (say) U class using the 2-4-1, and 2 more pax are travelling on a cash fare in the same cabin, the latter two will be booked into one of J, C, D, R or I classes - so they cannot be on the same booking as the U class passengers.
