As long as you don't straight out say that you want to cancel the card (just say you are thinking about it because the fee is too high, etc) but are transferred to retention, it can't hurt. Of course, there is always the chance they will mess up and cancel without you speaking it out specifically, but not very likely.
I recently inquired about replacing the paper edition of a magazine subscription with the digital version and instead of getting a response describing the costs and such I simply found out that my subscription was cancelled. Not a big deal for a magazine but that has me a little gun shy with my amex.
