(after winter 2018 remodel, no more smoking rooms)
Homewood Suites Riverport
have smoking rooms.
I know the first has an airport shuttle to/from STL, and I believe the second one does too.

Hyatt Regency St. Louis had smoking rooms last time I was there and even ashtrays in the hall. It appears from their website that smoking rooms are still offered.

The St. Louis Crowne Plaza downtown has smoking rooms

The Quality Inn in Florissant (very near STL) has smoking rooms:

I'm generally skeptical of Quality Inns, but a) this one has a really good rating on TripAdvisor, and b) I stayed in it a few years ago when it was a Hampton Inn (and it was just fine).
