If the conference is in West Midtown Penn Station allows for somewhat convenient mass transit options from JFK and also EWR. LGA has no rail service but is the closest and if you are taking a taxi should be the quickest, particularly to Midtown East.
LGA is closest and easiest. Car service on Elite or Dial7 can be called as you walk off the aircraft and will be pulling up as you walk out of the terminal (presuming carry-on only).
On another note...why is the question brought up so frequently? Just by looking at a map, it's clear that LGA is the closest to Midtown (closest = easiest to get to destination, in most cases). Sorry, it just kind of baffles me that this question comes up so often. I guess if you've never been there before....but...just look at a map!!!!

While helpful, a map of the NYC area doesn't tell the full story on which airport is the best for where you are going..too many factors to consider..time of day with traffic, reverse rush hour or not, where in Manhattan you are going, time needed to get to the city, cost, whether taking a taxi or train...too many variables for a map to answer...
LGA is my favorite by far. If you want to try to balance out cost/time, take a cab to Woodside LIRR, then LIRR into Penn Station. The cab fare should be in the $15-$20 range (with tip), and peak LIRR fare is $7.25 I believe. Even with moderate traffic in Queens, I've gotten from my arrival gate to Penn Station in under 30 minutes before. It's definitely a big time/cost saver during morning rush when everything into Manhattan is jammed.
I find JFK easiest to get to by public transit, but I despise the actual DL terminals there. That, and the lack of PreCheck. Obviously, that means I had to book a flight into JFK for next week...

I have always wondered why there was never a subway built from Grand Central to LGA airport....
Another vote for LGA for a quickie cab ride into the city. You can also make public transportation work from any of the three airports if you are being cost conscious (and LGA will be the cheapest in that respect as well).
If it is on the east side I would say LGA. West side it is more of a toss-up with JFK because Airtrain to LIRR or A train would be similar and I despise the M60 bus when going to the west side as it is ridiculously slow across 125th Street. Another thing to consider is just getting a cab to Astoria where you can catch the N or Q train (depending on time of day)
OP: use LGA, you won't regret it The only justification for not using LGA is if you REALLY don't want to take a cab.

Money, as said, and also the residents of the neighborhood it would run through (Astoria, in Queens) protested loudly the last time extending the N train was seriously discussed. It's dead, politically, even if the money showed up.

LGA will also take the longest by public transportation. JFK (cheap way: Air Train to E) is 1 hour into midtown. LIRR costs a little more, is a little faster into Penn Station.
