I am a long time lurker but I've only just signed up to take part in the discussions. There is an incredible wealth of information here and I would like to tap some of it if possible.
I am thinking of using a 2-4-1 voucher in September and I've narrowed my options down to Boston and New York. I am happy visiting either city but what route do you think I would benefit more from flying in First? Which airport is likely to have the best lounge experience? Which airport is the easiest to get to/from the centre of the city?

From NYC you have access to the CCR which is a nice lounge experience. BOS is a MF flight but don't let that put you off I had my best flight of 2012 on a flight in F to BOS last year.

I thought BOS was moving/had moved back to Worldwide fleet.
I would choose JFK - nearly guaranteed to get the new cabin, concorde room in both directions and I love New York!
New York has the CCR, but could you not fly to Boston, spend a few days there and then get the train to New York for a few days before flying out of JFK?

JFK wins on the lounge front but it has been a few years since i was in the BOS lounge.
Of course you could always book LON-JFK as LCY-JFK and then return JFK-LHR. But you could book a trip to BOS for while your there, in the grand scheme of things its not that far is it ?
on the way back you could book Y on AA BOS-JFK and thru check your bags, and still get access to the admirals club at BOS based on your onward flight ?

Personally, I wouldn't bother with F for such a short flight. save it for a properly long haul flight.
However, if you are going to do it then an Open Jaw is the way to go. 3-4 days in each with the train between (it's not worth the hassle flying NYC-BOS)
Both cities are much of a muchness to get to/from airports. Some sort of bus/transit to the metro and then slumming it with the great unwashed (hardly fitting if you've just Firsted in!) Get a cab, van or limo!
Both cities offer daytime flights back (why sleep through the first experience?) which IMHO is a great way to get over the jet lag. Besides it's often only a 6-6.5 hr flight back... not really enough for a decent kip.
Great train service on the Acela express between the two city centers so...
2 centre it. Fly into BOS and enjoy, train to NYC and be wowed, then back on the daylight maniac after a CCR brekky.
It really does help with the jet lag as you have an early start on the flight home, arrive at LHR early evening and then just get home and off to bed about 2, long lie in the next day. Job done. Done this several times on AC back from YYZ and it certainly beats the short sleep followed by '20 mins to landing' awakening and the inevitable groggy ratty day that follows.

I know folks say it's a "short flight" but Westbound to me it's an experience and it matters. Go for it.
+1 for NYC because of the city itself, flight options, probability of NF, CCR. The daytime flight JFK-LHR (morning departure) was EXCELLENT last summer - so much so that it's my next flight as well. I enjoy arriving in London and then heading to bed soon after.

NYC-BOS is only 30 minutes by air, so the flight times are ~7 hours to London in both cases. You cannot fly a longer flight on BA metal out of those cities, as they both land in London.
Either way, a 7 hour flight is a perfectly good use of a 2-4-1 in F.
That is a shame I found MF on this route truly excellent last year.
I think the day flights to and from JFK are an excellent way to experience F since you get more effective service time. Many longer routes in F are overnight (even flights of similar lengths like DXB) so you don't really get the same experience as most of the cabin is sleeping or trying to sleep.
Some of my best F experiences last year were on the relatively short day flight to CAI.
