Another vote for Tokyo.
There just are more convenient options.
As others have mentioned, April should be the cherry blossom season.
Geographically, Singapore and Hong Kong is very small. While greater Tokyo area is huge. You can easily fill a week in Tokyo while you would need creativity in the other two.
