Welcome to FlyerTalk, canoc_heou.
I am afraid upgrading on Cathay Pacific flights isn't that simple. You would first need to fly to earn the miles, and then the total accumulated miles you have earned over several flights can then be combined on an upgrade award.
You also cannot buy an upgrade. You can however buy a Business Class ticket.
You can join Asiamiles which is free, or any one of Cathay Pacific's airline partners who have a frequent flyer programme to earn miles. But Asiamiles is the only programme which you earn miles and redeem them for upgrades on Cathay Pacific and a few other airline partners. Information can be found on www.asiamiles.com
The Marco Polo programme is only worth it if you fly a lot and you want to earn elite level tiers. Information of the difference between MPC and Asiamiles can be found here: http://www.flyertalk.com/forum/11220622-post11.html

your miles are earned on the day that you fly, and not one the day that you buy your ticket, so yes, you can earn club miles for your flight in may. just present your marco polo number at check in and they will credit your miles.
you can upgrade to business using cash at any time. just call cx office. you can also upgrade on the day of your flight at the check in counter with cash, subject to availability. the cost will be the difference between the cost of the business class ticket and the economy ticket that you bought .

Cash upgrade - If you have a simple CX ticket, cash upgrade can be done anytime or at most airports if business seats are available on your flight. You have to pay the fare difference, which can be quite costly. If CX cannot figure out the fare from your ticket, then they may not be able to process a cash upgrade.
Mileage upgrade - You need miles in your Asia Miles account, at least enough so that you can top off. There are only limited seats, but CX may open many more seats closer to the departure date. If you do not have enough miles in Asia Miles, and if you have points in some of their partners such as SPG, you can transfer over. Note that transfer takes time.

I believe OP already joined MPC, which means he/she's automatically part of Asiamiles, no?

I believe he/she is. The Asia Miles no is the same as MPC no. Usually, pax will register for AM first and then MPC if they decide to pay the US$50 one-time registration fee.
