I'm trying to book a flight from ICN-MNL and it's showing as a 321 as below:

Does this mean that business class is available? When I try to book it, it doesn't work. If it is, what's the business like on that plane? It's > 4hrs, so want reasonable comfort like a domestic Air Canada flight would be in business...

Yes, "business class" service is sold on that flight. Equivalent to a domestic seat. Assuming inbound from overseas and then connecting at ICN (onwards to MNL), most pax sleep for much of the flight!

A321's Business class is more like domestic F/C seat as SFO_FT said. Nothing special - no sleeper, no PTV/AVOD. Just enjoy the meal service and have a good sleep!!

If you're lucky you could get the Ssam Bap (http://www.flyertalk.com/forum/showthread.php?t=691206). That's what I had the last time a flew on an OZ A321.
