Hello,
I understand Iberia has an interline baggage agreement with Singapore Airlines.
What if the Iberia flight is operated by Vueling? (flight has Iberia code)
Will the luggage still be checked through to final destination?

I really don't think so. Although their flights carry an IB code, they are clearly not Iberia flights and Vueling, as a LCC, probably doesn't have interline agreements with many airlines (perhaps only Iberia itself, if at all!).

Can't Vueling handover the luggage to Iberia, which in turn directly hands it over to Singapore airlines?
It would make sense to hand over the luggage to IB, as the ticket has IB flight numbers..

But the flight the passenger is travelling on has nothing (in operational terms) to do with IB.
Additionally, remember that even on connecting tickets, there can be circumstances where luggage can not be interlined, although almost everyone will do so on connecting tickets.
What you have got going for you: Vueling do understand the concept of connecting, they offer connecting flights themselves. They operate flights which have an IB codeshare, and IB certainly supports connections. [Although even if IB flight itself, I would not be surprised if they refused to through check bags to a non-oneworld alliance airline such as Singagopre airlines when you are not on a single ticket.]
Personally, I'd ask. But I wouldn't expect a positive answer.

In fact, VY does not have interline agreements with anyone (which is typical for an LCC).

Iberia only owns a minority stake in Vueling so would not expect too much!

I have done this many times on connecting Vueling flights as long as the agent knows the IB flight number as well as the Vueling one they can do it. If you have bought your ticket from Vueling they are not supposed to as Vueling charge for bags.
