Which is the best airliine and route to take from DC area to Venice? I found AirFrance to have more choices of time. Need suggestion.

This is a very broad question, what do you mean by best? Cheapest? Highest quality 1st class cabin? Limited # of connections?

There isn't a non-stop from DC to VCE, so no matter what you are changing planes. It all depends on where you would prefer to change, in the US or in Europe. If the US, then you can take either Delta or US Airways. If Europe, then there are many options, including Air France.

About a year ago, I found a very good connection flying United from IAD to BRU and then Brussels Air to VCE.

'Most direct' is still pretty difficult to define. Almost every European and many US airlines are going to be able to get you to VCE in one stop from DC, and for many of them the difference in mileage isn't that huge. I don't really think connecting in CDG, MUC, FRA, AMS, ZRH, LHR or FCO is going to make a huge difference in mileage. They're probably all within 300-400 miles flight miles of each other, which is <10% of the total flight.

forgot about delta from nyc. dca-nyc-vce. i avoid muc and fra and mil in winter. weather problems.

You should also think about two things: (1) what happens if your first flight is late and (2) how big is the airport/how difficult is the connection for you and your luggage. Unfortunately the answer to those two questions tends to prefer different routes. CDG is usually pretty good on the first question as there are likely to be multiple flights/day if your Paris flight is late, but there is also a chance that your luggage (if you check it) will not make the connection (as mine did not on my most recent trip through CDG to Italy).
