I am pretty sure I have seen this question answered here but I cannot find it and we are trying to wrap up our Uruguay arrangements. So I apologize and thank you in advance for the help.

We had hoped to spend the night in Colonia but can only spend the day there now and are considering taking the bus from Montevideo to Colonia and then the 5:30pm ferry back to BA on Sunday, Dec 4.

I tried to see if we could keep our car and drop it in Colonia but it doesn't seem possible - not sure if it is a problem because we would be dropping on a Sunday or if they just don't allow the one way rental.

We will each have a moderate size suitcase and carryon bag - is there any safe place to store in Colonia? If not, looks like we need to scratch this off our itinerary and take Buquebus from Montevideo to BA.

I haven't been in a few years, but the Buquebus port and/or bus station in Colonia are what I would check out first for storage options.

Another option may be to contact one of the cheaper family-owned/-run places and see if they would be willing to store your bag for the day or have a cheap "day room" (or even overnight room that you use for day use).

I went back on the Thrifty car rental site and was able to book the car for pickup Montevideo airport and drop Colonia downtown, for a $45 drop fee. We are happy campers now!
