Looking for singles scene with 40-50 year old single women. Not looking to go to clubs with 20-25 year old crowd. 70's music, etc.

No self-respecting woman of a certain age would be caught dead at a Barry Manilow concert. [I feel compelled to defend my generation though I'm a bit older than OP's target.]

Try the Ambhar Lounge at the "new" Trop. There is usually plenty of what you are looking for on the dance floor.

Stand in the middle of the Las Vegas Strip and throw a wiffle ball in any direction. Or go to the Rio and start playing the slot machine at the exit to Chippendales right before the show lets out.

Just hang out at the bar at the Las Vegas Hilton. Everyone there, (patrons, staff, hookers) is at least in their 50's...if not older.
