Not sure if I can from the terms and conditions for the new promo of 2 Stays for 1 free night.

There are multiple aspects to the new promo. One is:

"Members that qualify for the Promotion and pay for each of their qualified stays using a Best Western Rewards MasterCard® will receive 1,000 Best Western Rewards bonus points along with a Free Night"

At the very least, that part of the promo won't work if you use the gift card for those stays. (Of course, if you don't have a BW MC to begin with, you wouldn't have gotten those 1000 points anyhow. Plus, if you're elite, you can get the free night twice, but the 1000 bonus points only once, so in that case you could presumably use the $50 gift card for one of the second pair of stays.)

OTOH, I don't know whether BW can tell a gift card you bought from a gift card they gave you (once you go to use it). Assuming not, this 1000 points promo might be the only part you'd lose out on, as a stay paid with a gift card is still classified as a paid stay. (Furthermore, unless you are someplace where BWs are extroadinarily cheap, you're not likely to have the $50 gift card cover even an entire one night including taxes.)
