Taxi is the quickest way, but there might be a long taxi line. (Depends on luck). You could call a car service (212-777-7777). You can call them after you've picked up your bags and are about to leave the terminal. They will pick you up in a few minutes--less wait than a long taxi line. They'll tell you the exact place they will pick you up. It's outside the terminal right next to the taxi place. They'll pick you up without a reservation, though they always ask whether you have one, so I wonder if they give some sort of preference to people with reservations. (I've never made one). Good luck!

The fare to Manhattan is $46. Regular taxi fare is $45. If you go over the Triboro bridge or under the tunnel there's a charge for the toll. Since you'll be staying in midtown Manhattan you could go over the Queensboro bridge--it's free. (Upper level of the bridge is faster than lower level.) However there's sometimes nighttime construction on the bridge and drivers don't always want to go there. Then there's the tip--usually 15% or more. Basically the taxi and car service are the same price.

If you are carrying on take the AirTrain to its first stop and pick up a taxi from there it will be cheaper and quicker if there is a line at the taxi stand

Depending on the exact time you arrive, taxi may not be the best choice this weekend. They are doing construction on the FDR with full closures in the overnight hours. If you do a taxi, they may not be able to take the highway depending on the time, and will be forced to come in over the Queensboro which just crosses the FDR.

Closed from Midnight to 9 am, southbound 96th to 34th, northtbound the entire length from the Brooklyn to Triboro bridges. If leave before 11 or 11:15 it shouldn't matter, anything after that may.

You will probably totally miss it, you should be early enough, but just in case.....
